Water Quality and Metal Ions in Peptide Serum Stability

The laboratory used fresh deionized water. Production used a loop that had been idle over the weekend. The batch turned dull green and lost viscosity, yet the raw peptide was blamed. Water is the largest ingredient in most peptide serums, and small differences in hardness, trace metals, microbiology or conductivity can change color, preservation and rheology. A supplier COA cannot protect a formula from uncontrolled process water.

Define water by specification

Write chemical and microbiological limits appropriate to the process, with sampling points, frequency and test methods. Conductivity is useful but cannot reveal every contaminant. Include controls for system start-up, sanitization, stagnant branches and storage. Potable source compliance alone may not be adequate for a sensitive water-based serum.

Trace metals alter more than color

Microbiology starts upstream

A passing finished-product challenge test does not justify poor water-system control. Monitor microbial counts and investigate trends before they exceed limits. Biofilm fragments and endotoxin are not solved by adding more preservative to the formula. Define flush and sanitization procedures after idle periods.

Run production-water pilots

For scale-up, make a pilot with water from the actual system and process timing. Compare it with the laboratory control for pH, conductivity, color, viscosity and stability. This simple test often reveals a transfer problem before a 100-kilogram batch is filled.

Control changes and maintenance

Membrane replacement, sanitization chemistry, pipe modification or a new source supply can change water behavior. Route changes through quality review. Record point-of-use samples alongside commercial batch records so deviations can be reconstructed.

Investigate a discolored batch

Quarantine before correction. Compare water, raw-material retains, equipment contact, pH and oxygen exposure. Test trace metals if justified. Do not add chelator blindly to a copper peptide batch; first understand whether it will compete with the intended complex.

A useful water-system deviation plan

When point-of-use water misses a limit or trends unexpectedly, stop affected manufacture and identify the last acceptable sample. Map batches, cleaning solutions and intermediate stocks exposed during the interval. Resample according to a written investigation; do not test repeatedly until one passing result appears. Review sanitization, membrane pressure, conductivity, microbial counts and maintenance. If product was made, assess the actual formula and risk rather than assuming finished-product preservation erases the deviation. Record disposition and corrective action. For recurring color or viscosity differences, link water data to batch trends. That history can show whether the problem follows a loop branch, idle period or seasonal source change.
